Innovative Biotechnology for Generation of Cardiac Tissue

Heart transplantation remains the only curative treatment for end-stage heart failure. This life-saving option continues to be limited by low number of organ donors, graft rejection and adverse effects immunosuppressants. Engineering bioartificial hearts from acellular native-derived scaffolds stem cells has gained attention because its potential overcome these limitations. In this study, rat (n = 20) were decellularized means coronary perfusion with 1% sodium dodecyl sulfate (SDS) in a modified Langendorff device. The electrical field behavior SDS molecule was studied it assumed that when applying an alternating current, exposure time tissue detergent might decrease. To repopulate extracellular matrix (ECM), human mesenchymal (hMSCs) used, induced differentiate into cardiomyocytes (CMs) 5-azacytidine (5-aza). results showed no cellular debris intact ECM following decellularization. Decellularization presence electric proved faster, decreasing risk damage due detergent. After cell seeding culturing eight hMSCs, recellularization process analyzed using optic microscopy (OM), which suggestive CMs. study presents novel efficient decellularization protocol suggests hMSCs can useful generation heart.
